4- Hydroxyethyl-1- piperazineethanesulfonic acid (HEPES) is a biological buffer. It is used as reaction buffers, pre-hybridization buffers and hybridization buffers for separating and analyzing RNA nuclide components. It is used for 3'-end labeling of RNA with T4 RNA ligase. It is contained in biochemical diagnostic reagent kits, DNA/RNA extraction reagent kits and PCR diagnostic reagent kits. It has no cytotoxic effects on cells. It is a hydrogen ion buffer that can maintain a constant pH range for an extended period of time. The final concentration used is 10-50 mmol/L, and 20 mmol/L HEPES in the culture medium is generally sufficient for buffering capacity.
